APS Plasmagen Nitrate is a dietary supplement with a complex of ingredients including beta-alanine, citrulline malate, caffeine and theobromine.

Beta-alanine is an amino acid which is a precursor in the synthesis of carnosine – a compound which delays the onset of fatigue during training and facilitates more effective muscle function by activating myosin ATPase, an enzyme involved in the initiation of muscle contraction. 

Citrulline malate is a combination of the organic salt malate with an amino acid which contributes to better muscle recovery and pumped feeling during strength training.

Caffeine and theobromine are methylxanthines found in, among others, coffee and chocolate. Their effects involve reduced fatigue and pain and also improved cognitive functions.

Better blood flow and muscle growth
For long-term and healthy function, a muscle needs metabolic fuel and oxygen which are provided in blood vessels to every muscle cell. In order to be efficient, this process requires normal blood flow which is regulated by factors related to the nervous system, hormones and parathormone. One of these is nitrogen oxide (NO) which acts as a muscle relaxant – it causes the relaxation of smooth muscle and vasodilation that improve muscle perfusion.

NO synthesis takes place in the blood vessel endothelium synthesised from arginine directly by the enzyme called nitric oxide synthase. Citrulline is an intermediate metabolite which facilitates the synthesis of arginine and consequently NO.

Apart from vasorelaxation manifested by muscle pump, this amino acid has a protective effect on the endothelium of blood vessel cells, thereby delaying their pathological processes (among others, the accumulation of atherosclerotic plaque).

In addition, it may contribute, among others, to an increase in ATP production (by enhancing processes of glycogenolysis and glycolysis) required for muscle function. It has been also observed that it eases the blockade of the mTOR signalling pathway, consequently stimulating the biosynthesis of muscle proteins. What is more, as one of the intermediates in the urea cycle, citrulline may also help remove ammonia which inhibits aerobic metabolism (pyruvate -> acetyl-CoA) in muscle cells and thus hinders their efficient function due to ATP deficiency.

More effective recovery
Beta-alanine is a metabolite of cysteine formed from cytosine, carnosine and anserine and involved in the synthesis of two crucial compounds – coenzyme A as well as carnosine. One of these compounds is an important “activator” of acyl groups and that is why it may serve as substrates in the reaction of synthesis (e.g. of fatty acids) or combustion (e.g in pyruvate decarboxylation, resulting in the products of the Krebs cycle).

Secondly, carnosine contributes to an increase in the absorption of copper and may thus have a key impact on the activity of SOD (superoxide dismutase which comprises a copper and zinc atom) in the central nervous system. Consequently, it helps protect the body against harmful effects of free radicals (prevents from the development of Parkinson’s and Alzheimer’s disease). By affecting GABAA receptors, it may show similar activity to their antagonists.

In addition, carnosine acts as a buffer in skeletal muscles, thereby reducing acidosis (caused by intensive metabolism) which inhibits glycolysis and muscle cramps. Consequently, it improves their function.

Reduced feeling of fatigue and pain
Methylxanthines – caffeine and theobromine – are one of the most popular doping agents. In terms of their impact on mood, their most distinctive activity is that they are antagonists for adenosine receptors which results in reduced fatigue. Their effect on adenosine receptors is not negligible because the aforementioned antagonism contributes to increased secretion of serotonin, which has an analgesic effect. This may improve also adrenergic nervous transmission (stimulation), cholinergic transmission (in the cerebral cortex and hippocampus) and dopaminergic transmission (improved motor skills).

Caffeine and theobromine can also promote lipolysis via two mechanisms. In order to initiate it, there must be an increase in cAMP levels which contributes to the activation of the enzymatic apparatus required for “recycling” triglycerides. Methylxanthines prevents the breakdown of cAMP by inhibiting the activity of phosphodiesterase (an enzyme hydrolysing cAMP) and reducing the inhibition caused by adenosine, thereby blocking their receptors.

Alanine

Information


Alanine is an endogenous amino acid (i.e. having the ability to synthesize the human body other amino acids and pyruvate), building up protein. It has a key role in the transport of nitrogen between the muscles and the liver (i.e. Cahill cycle). It allows to transform glucose in non-sugar compounds. During prolonged exercise it enhances metabolism. It occurs in products of animal origin (meat, dairy, eggs) and vegetable origins (legumes, peas). It is used in the form of amino acid supplements in pre- and post-workout preparations. It complements the BCAA. Increased use of supplements with alanine is required in athletic training when high strength and endurance is required (e.g. weights lifting athletes marathon runners etc.).

Caffeine (1,3,7-trimethylxanthine, 1-methylotheobromine) is a purine alkaloid occurring in the natural state in plants (Coffea arabica, Camelia sinensis, Paulinia cupana, Ilex paraguariensis), which acts as a natural pesticide, acting toxic to insects. Peculiar smell and taste also deters herbivores. Caffeine in the body inhibits the adenosine receptors in the brain, fighting fatigue and allowing for action activation. It has a beneficial effect on the metabolism of oxygen, which increases the exercise capacity (strength and endurance) and rapid post-workout recovery. In the sports supplementation, caffeine is used as a stimulant nervous system. It has the thermogenic potential and in combination with other substances (e.g. Polyphenols, both occurring in coffee beans, such as cocoa beans, tea leaves and mate) can effectively participate in the reduction of body fat. It is included in many dietary stimulants (creatine stacks, no-boosters, beverages). It is a component of fat burners. It is also in the form of mono-preparation supplements.
Dosage: 100-600 mg per day. Due to the long release of activity should not be used in the evening, before
sleep.

Theobromine (3,7-Dimethylxanthine) is a purine alkaloid, poorly soluble in water, present in cocoa beans, as well as the leaves of yerba mate, tea, coffee, cola and peanuts.
