The absorption of the amino acid or its metabolites (HMB or HICA) takes place in the intestines owing to its transporter – glycoprotein CD98.

Providing 2-10 g of this compound helps reach the so-called leucine threshold. It is the minimal amount of leucine needed to improve the process of muscle protein synthesis. It depends from the activity of mTOR kinase.

Increased levels of leucine and its metabolites contributes to calcium retention inside the cell. The binding of ions of this element results in the activation of calmodulin which binds to the kinase hVPS34. This amino acid may also improve the body’s carbohydrate balance. This effect is achieved directly by leucine and consists in stimulating beta-pancreatic cells into releasing insuline. This contributes to more efficient utilisation of carbohydrates as a source of energy.

Since leucine activates sirtuins, it is believed to influence lifespan. Proteins from this group regulate the biogenesis of mitochondria and thus improve their function, prevent their death and increase energy levels. The total amount of mitochondria decreases over time, leading to a decrease in energy levels in the elderly.

The second amino acid which belongs to BCAAs is isoleucine. It is an isomer of leucine which, despite its similarity, shows a completely different effect. Its main task is to improve glucose uptake by muscle cells during physical activity and recovery. This contributes to maintaining high energy levels and protects glycogen from depletion. As a result, we achieve better recovery which is especially important to gym beginners and during hard training regimens. This effect is produced by the activation of PI3K/aPKC which does not depend from the activation of the mTOR kinase and AMPK.

Isoleucine promotes also anticatabolic conditions. This is due to the fact that it inhibits gluconeogenesis which consists in the breakdown of muscle proteins to obtain amino acids. In this process, they are a substrate for muscle glycogen resynthesis.

The last representative of BCAAs is valine. It is an amino acid which does not help improve composition of the body. However, valine protects it against catabolism because the body may use it to restore muscle glycogen in the case of hunger.

Isoleucine is an essential branched amino acid (BCAA), leucine-isomer.
